I've seen this a few times now, but I haven't been able to find a definition for it, or for what it might be derived from.

Here's an example:
今、オレホツとした?

should be from "hotto suru", with the small "tsu". "hotto suru" means "relieved, to be relieved".

well, here it is in another sentence:
座った時ホツとするような
カンジが似て

And, it kind of makes sense, but I've checked and re-checked, and I'm 98% certain that that's a big tsu (though it could be the typeface is throwing me off). Is there no other word it could be?

I think nhk9-san is right. I also suspect that there are certain publishers or magazines who print out the small "tsu" as big "tsu", not aware that not all readers are native-speaking Japanese. (It is a minor problem for Japanese.) This is because there had been other Japanese learners who have insisted it was so. I don't read manga anymore, so I can't tell which magazine. But this is probably the case with what you're reading.

カンジが似て:感じが似てー> "look alike (in general impression or atmosphere)"

And let's remember that it used to be standard practice to use the large "tsu" in places where modern works use a small "tsu". The distinction in reading/meaning is not new, but the distinction in publishing practice is definitely postwar.

I've always found that the small 'tsu' is very illusive when reading vertically. Probably a personal defect of mine, but maybe worth mentioning.

Ah, I see now. That's very confusing of them to do that. I considered it being a small tsu, but it threw me off that the ho was in katakana, while the to was in hiragana.
