〜から〜まで from ~ to ~
九州（きゅうしゅう）
事 is usually written in katakana when saying 〜ことにする
します is present tense or general statement.
As you have already decided it should be past tense しました which means completion.

Is there a better way to say the above sentence? and also how do you include the word especially/specifically.?

I think you also forgot を in yours...

No, you should not add を here...　For 行きたい the particle へ showing direction （to Nagasaki and Ooita) is already used.

特に長崎県と大分県と佐賀県へ行きたい.
You can add 特に before 行きたい but normally it can be put at the beginning of the setence.

The first day is 初日（しょにち） as Elizabeth indicated, or 一日目（いちにちめ）.
